Akhil (Nagarjuna) owns a call-cab service. He falls in love with a doctor, Siri Valli (Ayesha Takia). Soon Siri Valli falls in love with Akhil as well. There is a hi-tech robber called Sonu (Sonu Sood) who steals money in a big way. Sonu used to work for Siri Valli's father and so adopts her as his sister after their family fall into bad times.Incidentally, Akhil and Sonu are enemies. When Sonu realizes that Siri Valli is in deep love with his arch enemy Akhil, he asks her to put an end to their relationship and to never see him again. Siri Valli goes to Akhil's house to find out why Sonu and him are enemies. Akhil reveals his flash back:Akhil, Sasha (Anushka Shetty), and Sonu meet in Mumbai in their needy days as unemployed graduates. They soon become friends as they all struggle to find work. Sasha is Sonu's sister. Sasha suddenly develops pains in her stomach and needs an appendix operation. Sonu and Akhil don't have enough money. After a few days, they were forced to steal money. Then they decide to rob the money and make a living out of it. Sasha loves Akhil. But Akhil does not have any feelings for her. He treats her just like a friend. Akhil, Sasha, and Sonu are given one more task: to rob diamonds. After this one robbery, they can be settled for life. Late that same night at the biker's club, Sasha says openly that she will kill herself if Akhil does not love her. Being stubborn, Sasha decides to stay at the Biker's club very late. Meanwhile, she hears some men talking and finds out that it was the same men who told them to rob the diamonds. But the goons plan to kill Akhil, Sonu, and Sasha right after they succeed in the robbery. Sasha overhears them and the goons chase Sasha. They kill her and make it to believe that she committed suicide.From then on Sonu hates Akhil. Because Akhil couldn't love Sasha, Sonu hates Akhil. Still, neither Sonu or Akhil know that Sasha was killed by goons. After several incidents, they find out who really is behind Sasha's murder: Mama. Mama kidnaps Siri Valli and in ransom, tells Akhil and Sonu to rob the same diamonds they were supposed to rob in Mumbai. They successfully rob the diamonds by blowing up underneath the truck carrying the diamonds (inspired by The Italian Job) and falling into the sea. Akhil then goes into the ocean and gets the diamonds. The police start chasing Sonu and Akhil on motorboats. One of the police shoots Sonu and he falls into the ocean. Still, Akhil does not stop and he heads on over to Mama's place. Akhil gets a hold of Sonu, who faked his death, and Sonu rushes to get his sister from the clutches of the bad guys. Akhil and Sonu manages to kill everyone there and keep the diamonds.

Meanwhile, Shankar Narayana is alive and returns from coma, but still people think he is dead.His son creates that kind of world where his father is kept happy by using Brahmanandam and others. The movie takes interesting plot post interval as the main plot is revealed to story. Nayak is not only a mission for Ajay but is much more in case of a revenge. How is Mallesh (Kota Srinivas Rao), Meka(Sayaji Shinde) and Supreet are involved in betrayal, what is Ajay's actual plan. Will Ajay kill Nayak or Shankar Kills nayak. Will Shankar know the true world. Will Ajay marry Prashanthi.

Aishwarya (Ileana D'Cruz) is the daughter of central union home minister Raya (Prabhu Ganesan). Due to some reason, he doesn't allow his daughter to go on a trip with friends. She nonetheless goes to the trip. She then meets a guide, Shakti (Jr. NTR), who takes the group on a trip to several places. At first, she hates him because he uses every chance to get money from her. But when he saves her, she starts liking him. Afterwards, a few people who are chasing her finally find her in Kashmir and try to kill Shakti as he hurt'd them earlier. Their plan fails as Shakti turns out to be a Secret Agent and defends himself and catches the main person in this plan. When Aishwarya goes back to her father, he forgives her and asks whether she had taken a box in which there is a diamond. When she replies in the negative, he says that it is more important than his own daughter. Later, when Shakti goes to a temple, the diamond box falls off a basket his mother brought with flowers on top of it. They inform Mahadevaya who asks them to immediately come to Humpy with the box and Aishwarya. Their travel is blocked by a strong man who is then defeated by Shakti. He then kills one more person who tries to grab the box. Then Mahadeva Raya and a Swami (Nassar) come to him and then the flashback is revealed.It is said that Mahadeva Raya'a father (S. P. Balasubramaniam) was a royal king who was extremely generous and had the blessings of god which gave him the power that whenever he touched the diamond, it used to shine brightly emitting light. He reveals that apart from the 18 Shakti petas, there is a secret one which protects India and only their family knows of it. There is also a protector who guards it at all times. His name is Rudra (Jr. NTR). After every 27 years, they must go to the shakti peta where the protector will perform several spiritual rites which must be compulsorily performed. When he starts performing them, an Egyptian king who knows of this comes to destroy the shakti peta. The protector kills the king but a person who works under the Mahadeva Raya's father, Janaki Varma (Jackie Shroff) cheats him and kills both the protector and Mahadeva Raya as he sought money from the king. He also steals a sword (Maha Trishulam) which is necessary for the rites to be performed. When the protector is killed, his wife, at the same time, delivers a baby boy whom Rudra gives to his loyal servant. The servant tries his best to save the boy but circumstances force him to throw the boy into the river. The boy is then found by a childless couple who adopt him. After finding out his past, he sets out for revenge.Shakti then goes to Janaki's country where he is presently the underworld don and a multimillionaire. He kills Janaki and gets the sword. Finally he goes to perform the rites but is stopped by the Egyptian queen and her powerful son who comes back and cannot be defeated. The swami then tells him that the strong man can only be defeated by damaging his eyes. He then kills him and performs the rites.

Karan (Sonu Sood) loves Sneha (Tanushree Dutta), but is too much of an introvert to ever express his feelings to her.
